The Liverpool forward is thought to have ruptured his Achilles' tendon.
Coach Marco van Basten named defender Khalid Boulahrouz of Spanish outfit Sevilla as the youngster's replacement.
Holland - who won the tournament 20 years ago - open their Euro 2008 campaign against Italy on June 9 and will play France four days later.
